Heat shock (HS) response is well characterized by its rapid activation upon heat stress exposure and inactivation after stress. By examine transcriptomic alteration after HS, we have revealed a transcriptional reprograming (post-HS response) that differs significantly from the well-studied HS response in C. elegans. We also found that the post-HS and HS responses are differentially regulated and could be uncoupled from each other. Loss of post-HS response dose not impair survival of animals upon HS insult but rather abolishes long-term influence of a transient HS. One important question is that how cells coordinate the sequential arrangement of these two distinct transcriptomic alterations during and after HS. In the international CIBSS symposium we will present our latest work addressing temporal control of transcriptome by the small RNA pathway and how small RNA binding argonaut proteins interfere with RNA polymerase II to precisely determine acute stress response and subsequent long-lasting post-stress response.
